| I. Background | |

TL1A 又稱 TNFSF15，是 TNF 超家族的成員，在單核細(xì)胞、巨噬細(xì)胞、樹突狀細(xì)胞、 T 細(xì)胞等免疫細(xì)胞和非免疫細(xì)胞（如滑膜成纖維細(xì)胞、內(nèi)皮細(xì)胞）中均有表達(dá)。TL1A 是一 種 Ⅱ 型跨膜蛋白，通過(guò)與 TNF 同源結(jié)構(gòu)域 (THD) 相互作用自組裝成穩(wěn)定的三聚體。它主 要以膜結(jié)合形式表達(dá)，并形成穩(wěn)定的三聚體?？扇苄?TL1A (sTL1A) 是通過(guò)可變剪接或 TNF-α轉(zhuǎn)換酶 (TACE) 裂解產(chǎn)生的。
TL1A 競(jìng)爭(zhēng)性地與死亡受體 3（DR3）或誘餌受體 3（DcR3）結(jié)合，為下游信號(hào)通路提 供刺激信號(hào)，進(jìn)而調(diào)控效應(yīng)細(xì)胞的增殖、活化、凋亡及細(xì)胞因子、趨化因子的產(chǎn)生。 |
