| I. Background | |
| CD74 是一種具有多種免疫功能的多態(tài)糖蛋白，功能是其對 MHC Ⅱ類分子在抗原提呈 細胞的轉運的調(diào)節(jié)作用。同時 CD74 是巨噬細胞移動抑制因子（MIF）的受體復合物的不 可缺少的一部分，MIF 作為一種多功能的細胞因子樣分子，不但調(diào)節(jié)先天性和獲得性免疫 能力，而且在慢性炎癥性疾病和致癌方面也起到很大的作用。CD74 可以通過 Src 激酶激活 PI3K-AKT 信號通路進而調(diào)控下游的凋亡調(diào)控蛋白。PI3K-AKT 通路可以介導促凋亡蛋 白 BAX 和 BAD 的磷酸化失活，也能通過激活 NF-κB 的方式，促進 BCL-2 和 BCL-xL 的 表達，減少細胞凋亡，促進細胞存活。CD74 除了在機體免疫反應中發(fā)揮作用，在前列腺 癌、肝癌、肺癌、乳腺癌、黑色素瘤等多種惡性腫瘤細胞中的表達上調(diào)，并顯示其在腫瘤 發(fā)生、血管生成和增加腫瘤細胞的侵襲能力等方面存在重要作用。 | |
