| I. Background | |
| Programmed Cell Death Protein 1 （PD-1），一種在激活的 T 細胞上表達的受體，與其 配體 PD-L1 和 PD-L2 結(jié)合，負向調(diào)節(jié)免疫反應。 PD-1 配體存在于大多數(shù)癌癥中，PD-1： PD-L1/2 相互作用會抑制 T 細胞活性，并使癌細胞逃避免疫監(jiān)視。 PD1/PDL1 信號轉(zhuǎn)導通 路是腫瘤免疫抑制的重要組成部分，可以抑制 T 淋巴細胞的興奮，增強腫瘤細胞的免疫耐 受，從而實現(xiàn)腫瘤免疫逃逸。PD1 與 PDL1 結(jié)合可以減弱 T 細胞介導的免疫監(jiān)視，導致免 疫反應缺失，甚至導致 T 細胞凋亡。PD1/PDL1 抑制劑可解除抗腫瘤 T 細胞的免疫抑制， 從而導致 T 細胞增殖并滲透到腫瘤微環(huán)境中并誘導抗腫瘤反應。PD-1：PD-L1/2 通路還參 與調(diào)節(jié)自身免疫反應，使這些蛋白質(zhì)成為多種癌癥以及多發(fā)性硬化癥、關節(jié)炎、狼瘡和 I 型糖尿病的有希望的治療靶點。酪氨酸磷酸酶 SHP2 是 T 細胞功能的關鍵調(diào)節(jié)因子，它介 導 TCR 下游的激活信號以及 PD-1 下游的抑制信號。 | |
