血小板衍生生長(zhǎng)因子(PDGF)是一類重要的生長(zhǎng)因子,主要由血小板、內(nèi)皮細(xì)胞和 巨噬細(xì)胞分泌,在細(xì)胞增殖、遷移、分化和血管生成等生物學(xué)過(guò)程中發(fā)揮關(guān)鍵作用。它不 僅在正常生理過(guò)程中(如傷口愈合和組織修復(fù))起重要作用,還在多種病理過(guò)程中(如癌 癥、纖維化和動(dòng)脈粥樣硬化)扮演重要角色。PDGF 家族包括 PDGF-A、PDGF-B、PDGF- C 和 PDGF-D 四種亞型。PDGF 通過(guò)與其受體(PDGFR)結(jié)合,激活下游信號(hào)通路,調(diào)控 細(xì)胞行為。 血小板衍生生長(zhǎng)因子受 0 體α(PDGFRA),又稱為 CD140A 或 PDGFR2,是一種細(xì)胞 表面酪氨酸激酶受體,屬于血小板衍生生長(zhǎng)因子家族。PDGFRA 與配體 PDGF 結(jié)合后會(huì)二 聚化以及磷酸化,激活包括 JAK-STAT3、PI3K-AKT-mTOR 和 RAS-MAPK 等下游信號(hào)通路, 在調(diào)節(jié)關(guān)鍵細(xì)胞功能如增殖和凋亡方面有重要作用。PDGFRA 蛋白被激活異常時(shí),則會(huì)導(dǎo) 致腫瘤的發(fā)生并促進(jìn)腫瘤血管生成。 |
